Posting Summary
Rutgers, The State University of New Jersey is seeking a Postdoctoral Fellow for the Department of Microbiology, Biochemistry and Molecular Genetics within the New Jersey Medical School. A postdoctoral fellow position is available in the Petrou lab at Rutgers-New Jersey Medical School (Newark, NJ). Our research focuses on the structural biology of membrane proteins using single particle cryo-electron microscopy (cryoEM). Our goal is to characterize the structure and function of bacterial membrane enzymes involved in antibiotic resistance, and eukaryotic receptors and enzymes relevant to human physiology and pathology.
The Petrou Lab is located in Newark, New Jersey, 20 min away from New York city. The Rutgers New Jersey CryoEM/ET facility, located in Piscataway, New Jersey, provides access to cryoEM and cryoET instrumentation including a 200kV Talos Arctica microscope, an Aquilos 2 FIB-SEM microscope, and a Leica cryo-CLEM setup. In addition, a local cryoEM facility at NJMS that will house a 100kV Tundra microscope equipped with speed-enhanced Ceta camera, and sample preparation equipment, will begin operating in the next few months, and is located in the same building as the Petrou Lab.
The Postdoctoral Fellow will be responsible for the expression, purification and structure determination by single-particle cryoEM of membrane proteins relevant to polymyxin antibiotic resistance. In addition, the Postdoctoral Fellow will be responsible for biophysical or biochemical characterization of target proteins, including protein-ligand or protein-protein interactions. Moreover, the Postdoctoral Fellow will assist the PI in lab organization, daily operations, personnel training, and grant writing activities.
The successful candidate will have a Ph.D. in Structural Biology, Molecular Biology, Biochemistry, or related fields. Preference will be given to candidates with experience in membrane protein expression and purification and/or single-particle cryoEM. Good communication & organizational skills and the ability to work independently are important for this position. Salary will be commensurate to the candidate’s experience and qualifications and based on the Rutgers postdoc salary schedule.

Among the key duties of this position are the following:
  • Performs the following research-related tasks:
  • a. Protein expression and purification – overexpression using bacterial, insect or mammalian systems, SDS-PAGE, and chromatographic separation using FPLC and HPLC.
    b. Biochemical characterization of recombinant and native proteins (Western blotting, immunoprecipitation, cross-linking, etc.)
    c. Molecular biology (expression vector design and construction, PCR, site-directed mutagenesis, in vitro transcription, etc.)
    d. Biophysical characterization of recombinant or native proteins and their cognate targets (i.e. ligands or proteins) using Microscale thermophoresis (MST).
    e. Sample preparation for single-particle cryoEM using cryo-plunging equipment (i.e. Vitrobot or equivalent).
    f. Screening and data collection of single-particle samples using the Tundra or Arctica transmission electron microscopes.
    g. Determination of three-dimensional structures of proteins using cryo-electron microscopy data and appropriate computational programs/resources (e.g. Relion, cryoSPARC, etc.).
  • Collects, compiles and presents data with appropriate conclusions, recommendations, etc. pertinent to research activity.
  • Communicates regularly with the Principal Investigator.
  • Maintains detailed and accurate experimental records.
  • Participates and presents in journal clubs, seminars and conferences.
  • Prepares reports for peer-reviewed scientific publications, and contributes to grant writing.
  • Performs training of lab personnel in molecular biology, biochemistry and single-particle cryoEM.
  • Supports the daily operations within the lab, e.g., lab reagent generation and administrative tasks.
  • Performs laboratory inventory monitoring and ordering.
